oh you give it to the old lady in town and she will use the rune to give you more abilities...
i havent plaied for a while so i forgot what runes do what...
For example: i remember a kind of rune that will give you 7+ hp or something...

...i dont see any spam. this post is fine.

@cannibal: go to jezebelle's runes in d3jsp hall and you can either use one rune at a time or use a rune combo. the effects last untill your next dragon kill (when you are reset to level one with no buffs except for any extra hp that you might have earned other than from leveling)
